Polygynax vaginal capsules are a combined local anti-infective medicine containing nystatin, neomycin sulfate and polymyxin B sulfate. Neomycin and polymyxin B are antibiotics acting against selected susceptible bacteria, while nystatin is an antifungal medicine active against susceptible Candida species.
The medicine is intended only for adult women and is used for the local treatment of vaginal infections caused by microorganisms sensitive to its active ingredients. Polygynax is supplied only with a medical prescription.
Changed vaginal discharge, itching, burning or an unpleasant odour do not by themselves confirm that Polygynax is the correct treatment. Bacterial vaginosis, candidiasis, mixed infection, irritation and sexually transmitted infections may cause similar symptoms but require different management.
Do not use leftover capsules from an earlier treatment. Excessive or prolonged use may increase the risk of resistance, superinfection, allergic eczema and systemic adverse effects.
Polygynax – approved uses, application and precautions
What is Polygynax used for?
- bacterial vaginitis;
- vulvovaginitis caused by susceptible Candida albicans or non-albicans species;
- mixed bacterial and fungal vaginitis;
- bacterial vaginosis.
Official recommendations for the rational use of antibacterial medicines must be followed. Polygynax should not be used preventively or for every episode of vaginal discomfort without medical assessment.
How is Polygynax used?
| Users | Recommended dose | Duration |
|---|---|---|
| Adult women | One vaginal capsule in the evening | 12 consecutive days, as prescribed |
| Children and adolescents | Not intended for use | Approved only for adults |
Insert one capsule deeply into the vagina in the evening before bedtime, preferably while lying on your back with your legs bent and slightly apart.
Wash your hands before and after use. The capsule is intended only for vaginal administration and must not be swallowed.
Practical advice during treatment
- Use your own towel to reduce the risk of contamination.
- Wear breathable cotton underwear.
- Avoid vaginal douching.
- Do not use tampons during treatment.
- Do not interrupt treatment during menstruation.
- Do not extend treatment beyond the period prescribed.
- The doctor will decide whether the partner also requires assessment or treatment.
Do not apply two capsules after a missed dose. Continue with the next dose at the usual time.
Condoms, diaphragms and spermicides
Polygynax is compatible with male condoms made from latex or polyisoprene.
It must not be used with:
- polyurethane male condoms;
- female condoms;
- diaphragms.
Combined use with local spermicides is not recommended because vaginal treatment may reduce their contraceptive effectiveness.
When must Polygynax not be used?
- allergy to nystatin, neomycin, polymyxin B or any excipient;
- allergy to related aminoglycoside antibiotics, polypeptide antibiotics or polyene antifungals;
- allergy to peanuts or soya, because the capsules contain hydrogenated soya oil;
- use of polyurethane male condoms, female condoms or a diaphragm;
- children or adolescents;
- without medical assessment when the cause of the symptoms is unknown.
When is particular caution required?
Tell the doctor if you have impaired kidney function. Systemic absorption of neomycin and polymyxin B through the vaginal mucosa cannot be completely excluded, particularly with excessive or prolonged use.
Stop treatment and contact a doctor if burning, itching, swelling or redness becomes worse following application.
Local sensitisation to an antibiotic may make future treatment with the same or a related antibiotic more difficult.
Pregnancy and breastfeeding
Use during pregnancy is not recommended unless a doctor considers the treatment necessary. Neomycin is an aminoglycoside antibiotic with a potential risk of ototoxicity if systemically absorbed.
Use during breastfeeding is also not recommended unless the doctor decides otherwise after assessing the benefits and risks.
Possible side effects
The frequency of reported reactions is not known. Possible effects include:
- rash, itching or hives;
- burning or stinging at the application site;
- irritation, redness or swelling;
- a serious allergic reaction, including anaphylaxis.
- difficulty breathing or swallowing;
- swelling of the face, lips, tongue or throat;
- widespread hives or a rapidly developing rash;
- severe swelling, pain or intense burning after application;
- unexpected vaginal bleeding;
- severe lower abdominal pain, fever or marked weakness;
- rapid worsening of symptoms.
Excessive or prolonged use
Excessive or long-term use may cause systemic effects on hearing or kidney function, particularly in patients with kidney impairment. It may also increase the risk of allergic eczema, resistant microorganisms and superinfection.

Avoid vaginal douching, tampons, aggressive intimate washes and perfumed products during treatment.
Oral and vaginal probiotics are not substitutes for Polygynax. Their timing in relation to local antibiotic treatment should be discussed with a gynaecologist or pharmacist.
