Tregona 5 mg and 10 mg contains donepezil hydrochloride, an acetylcholinesterase inhibitor used for the symptomatic treatment of mild to moderately severe Alzheimer’s dementia in adults.
Donepezil increases the availability of acetylcholine in the brain by slowing its breakdown. It may temporarily support memory, thinking and everyday functioning in some patients, but it does not cure Alzheimer’s disease or reliably stop its progression.

Tregona – what is it used for and how should it be taken?
Treatment must be started and supervised by a doctor experienced in diagnosing and treating Alzheimer’s dementia. It should be started only when a caregiver is available to monitor regular medicine use.
The usual initial dose is 5 mg once daily for at least one month. After clinical assessment, the doctor may increase the dose to 10 mg once daily. The maximum recommended dose is 10 mg daily.
- Take the tablet in the evening immediately before going to bed.
- Swallow it whole with sufficient water.
- Food does not significantly affect its action.
- Do not take more than the prescribed single daily tablet.
- Do not change the strength or stop treatment independently.
- Regular medical reviews are required to assess continued benefit.
If a dose is missed, skip it and take the next dose at the usual time. Do not take a double dose. Contact the doctor before restarting if treatment has been missed for more than seven days.
Tregona may slow the heart rate and cause dizziness or fainting. A fall, loss of consciousness or a new markedly slow or irregular pulse requires medical assessment.
Treatment should continue only while the doctor considers that the patient is receiving a meaningful clinical benefit.
Tell the doctor about a history of stomach or duodenal ulcer, gastrointestinal bleeding, seizures, heart-rhythm or conduction disorders, prolonged QT interval, low potassium or magnesium, asthma, lung disease, liver disease or difficulty urinating.
Important interactions
Interactions may occur with medicines affecting heart rhythm, antidepressants, antipsychotics, macrolide and fluoroquinolone antibiotics, ketoconazole, rifampicin, carbamazepine, phenytoin, galantamine, anticholinergic medicines, beta-blockers, NSAID painkillers, muscle relaxants and anaesthetics.
Avoid alcohol. Tell the surgeon and anaesthetist that the patient is taking donepezil before any procedure requiring anaesthesia.
Possible side effects
Very common effects are diarrhoea, nausea and headache. Other possible effects include vomiting, loss of appetite, muscle cramps, tiredness, insomnia, nightmares, hallucinations, agitation, aggressive behaviour, dizziness, fainting, rash, itching, urinary incontinence and an increased risk of falls.
- black stools or fresh blood in the stool;
- seizures;
- fainting or a markedly slow or irregular heartbeat;
- high temperature, severe muscle stiffness, sweating and altered consciousness;
- severe muscle pain or weakness with fever or dark urine;
- difficulty breathing or swelling of the face, lips, tongue or throat;
- new uncontrolled movements of the face, tongue or limbs.
A caregiver should monitor each daily dose and prevent accidental repeat dosing. Monitor pulse, dizziness, fainting, appetite, persistent diarrhoea, vomiting and unintended weight loss.
Do not independently introduce ginkgo, bacopa or other memory products without checking the complete treatment because some ingredients may affect bleeding, heart rhythm or seizure risk.
